dTAG-13 is a heterobifunctional PROTAC research reagent for rapid, controlled degradation of proteins fused to the engineered FKBP12^F36V^ tag. One end of the molecule binds FKBP12^F36V^; the other recruits the cereblon (CRBN) E3 ubiquitin ligase complex. This brings the tagged protein to the ubiquitin–proteasome pathway for degradation. By introducing the FKBP12^F36V^ tag to a protein of interest, researchers can use dTAG-13 to study the effects of acute protein loss. Applications include target validation, pathway analysis, and studies of protein function in cells. The protein must carry the FKBP12^F36V^ tag for the intended dTAG response; dTAG-13 is not a general degrader of untagged FKBP12.
